Purpose:
|
Retinopathy of Prematurity(ROP) is a major cause of blindness in premature newborns worldwide. Hyperoxia is one of the most important risk factors in the development of ROP. Hyperoxia leads to suppression of Hypoxia-Inducible Factor (HIF)-1 and vascular endothelial growth factor (VEGF), which in turn causes both cessation of retinal vessel growth and loss of some existing retinal vessels (phase 1 ROP). We hypothesized that inhibition of Phosphodiesterase 5 enzyme suppress retinal vasoconstriction secondary to hyperoxia and prevent ROP phase 1 and subsequently ROP phase 2

Methods:

0ne hundred newborns with respiratory distress syndrome treated with oxygen were randomized into two groups as Group A, Placebo (as control group) and Group B (intervention group), sildenafil were administered via nasogastric tube. Occurrence of ROP phase 1 as primary outcome and stage 2-5 ROP, duration of mechanical ventilation, Nasal Continuous Positive Airway Pressure (NCPAP), oxygen therapy and duration of hospitalization as secondary outcomes were assessed

Results:

Fifty patients in each group were studied. ROP phase 1 was seen in 11(22%) and 7(14%) of placebo and interventional group, respectively
|
Conclusion:
|
The current study showed that sildenafil might prevent neovascularization in premature infants.
